The work in the Boekelheide laboratory examines fundamental molecular mechanisms by which environmental and occupational toxicants induce injury. Current projects include the development of novel in vitro approaches to safety assessment, use of xenotransplantation approaches for human-relevant toxicity testing, and the discovery of sperm molecular biomarkers that reflect testicular injury.

Biography

Kim Boekelheide is Professor of Pathology and Laboratory Medicine at the Brown University School of Medicine. He received his B.A. from Harvard University, and M.D. and Ph.D. from Duke University. His research examines fundamental molecular mechanisms by which environmental and occupational toxicants induce testicular injury. Current projects include the development of novel in vitro approaches to safety assessment, use of xenotransplantation approaches for human-relevant toxicity testing, and the discovery of sperm molecular biomarkers that reflect testicular injury. He is Director of the Brown University Superfund Research Program and Children's Environmental Health Center. His research has been continuously funded by the National Institute of Environmental Health Sciences since 1985 and he has received several awards including a Burroughs Wellcome Toxicology Scholar Award (1994-1999).

Research Description

My career has been devoted to understanding how environmental and occupational chemicals affect mammals at the biochemical, cellular, and molecular level. In the laboraory, much of this effort has been devoted to studying how the mammalian testis functions and the underlying mechanisms responsible for toxicant-induced testicular dysfunction. Hormones, paracrine signaling factors, signal transduction processes, and intracellular response pathways are the key cellular and molecular regulators of this wonderfully complex system responsible for spermatogenesis.
More broadly, as a member of the National Academy of Sciences committee that produced the report entitled "Toxicity Testing in the Twenty-first Century: A Vision and a Strategy," I have thought long and hard about toxicity testing, and I know that a transformative change is inevitable. We currently rely on 20th century technologies that are too slow, too expensive, and fail to take advantage of the ongoing revolution in biology. With the new technologies and bioinformatics tools now being developed, we can shift the paradigm of toxicity testing, characterize the responsible toxicity pathways, and develop a robust mechanistic explanation of toxicant action.
As Director of the Children's Environmental health Center at Brown University, I have advocated the use of human tissue xenotransplantation approaches to enhance the human relevance of our research effort, which is focused on understanding the fetal basis of later life diseases. This has involved intense investigation of epigenetic responses to toxicant exposure.
As Director of the Superfund research Program at Brown University, I have strongly promoted the value of developing academic-government-community partnerships to enhance the value and relevance of our work. My project in the Superfund addresses deficiencies in the current methods used to monitor environmental and occupational exposures that target testicular function which rely on serum hormones and semen parameters (sperm count, motility and morphology). These traditional assessments of testicular function are insensitive and highly variable. My project identifies sperm molecular biomarkers by exploring fundamental processes in spermatogenesis and learning how to detect the fingerprints of their disruption in the most robust manner possible. The approach involves an exciting interdisciplinary integration of basic biology and translational science, ferreting out the consequences of toxic exposures by applying newly discovered knowledge to protect human health.
